The present study was aimed to investigate the β-adrenergic antagonistic effect of lobeline on the receptor and effector transduction pathway. Lobeline, a narcotic alkaloid with antidepressant and anti-anxiety effects, was used as a positive control drug. In this study, the isolated aorta was precontracted with phenylephrine. Aortic rings were contracted with phenylephrine and treated with phenylephrine and different concentrations of lobeline. The following physiological parameters were recorded: 1) lumen diameter; 2) maximal tension; 3) lumen diameter to tension ratio. The phosphorylation and activation of G-proteins were investigated using Western blotting. Phosphorylation of G-proteins and phosphorylation of PKA was quantified by scanning densitometry. Ca2+ influx was quantified by an atomic absorption spectrophotometer. The results showed that lobeline markedly antagonized the contractile response induced by phenylephrine.
